Museum doors open at 8am. Tour description commences in line pre-entrance inc. full explanation of the Sistine Chapel. You will be in the first 5 groups entering the museums. Approximately 300 groups enter the Vatican Museums every day. By the time you enter the Sistine Chapel, there will be approximately 50 people inside too.
